Tolerance of sonidegib after intolerance of vismodegib—Experience in two patients with nevoid basal cell carcinoma syndrome (Gorlin syndrome)

Nevoid basal cell carcinoma syndrome (NBCCS, Gorlin syndrome) is a rare genetic condition characterized by the early development of numerous cutaneous carcinomas (BCCs). Although most BCCs are surgically treated with total resection, some lesions may proceed to locally advanced or metastatic stage. Systemic treatment hedgehog inhibitors (HHIs) such as Vismodegib Sonidegib indicated in this population. We report cases two patients confirmed diagnoses NBCCS. Both had undergone multiple surgical excisions and been oral 150 mg/day for tumour. They both discontinued therapy due its specific adverse effects (AEs) now being Sonidegib, which has better tolerability complete response. The aims was demonstrate efficacy HHI NCBBS who previously but it because AEs. Our experience shows that can be considered intolerant responding Vismodegib.
